Yes, it is possible to make 3-way calls on a Polycom VVX 250. Here's how you can do it:
Start the first call: Dial the first number and wait for the call to connect.
Add a second call: Once the first call is connected, press the "More" softkey, and then select "Conference". This puts the first caller on hold. Now, dial the second number and wait for it to connect.
Join the calls: Once the second call is connected, press the "More" softkey again, then press "Conference" to merge both calls into a 3-way conference.
All participants are now connected in a 3-way call.
